The first time I heard it emphasized was in "Babylon 5"..

"Who are you?" was the question the Vorlons liked to ask.. "What do you want?" is the one the Shadows asked. They weren't easy questions as the askees found out.. in fact, two characters found the question "Who Are You?" quite.. shockingly difficult... The phrase "Who are you? What do you want?" was spoken in whole in the final episode of Babylon 5 by Lorien, the first of all the sentient beings. To wit, those were 2 very central questions to the story.

But then tonight I also saw it in a movie that came even earlier than that.. "Ghost in the Machine", a cyberpunk/horror(wannabe) movie.
