Anal penetration isn't painful when it's done correctly. If it's painful, you're not using enough lube, the receiver isn't sufficiently aroused or relaxed, or you're trying to do too much in terms of size and/or penetration too quickly.

But penetration isn't the topic of this thread. The OP said he wants to use his mouth on his wife's anus, not insert anything. If he does want to insert something at some point, he needs to talk to her about it beforehand or wait until she suggests it; otherwise, it IS likely to be a painful experience in some way.
