Well, I'll tell you a story. It was only a few months ago. There was this pretty hot chick I worked with. I knew right from the start that she really really wanted to fuck me. From my perspective I would rather have small hints that lead up to big hints instead of a girl being blunt and saying "Fuck me now" if we had never slept together.

I have to disagree with you there - some guys are easy. For a guy like me, who's already had to initiate any show of interest, a girl coming up to me and telling me she wanted me to fuck her would instantly cause a red flag. Not because I didn't believe her, but because there's no precedent for me being that lucky.

I think it really does depend on the guy. If he seems more confident and self-assured, direct approach would probably work. For the more shy and withdrawn, get them alone first (someone mentioned inviting them to your place for a friendly drink - that'd work), then make your move. In my experience at least, the shier types are much more responsive in private, one-on-one situations than in an open public setting.
