Best way to get a book published these days?

You're most direct route is self-publishing. If you want to publish through traditional routes then you have two choices. You can pray for an angel, or you can do a hell of a lot of work to sell your story.
 
You can use Draft2Digital, which will put it (for a percentage of the price) on all the sites (even Amazon, but my pub puts it there himself). There are mom-and-pop outfits out there, and small publishers. You can submit your story, and many will accept it. Don't use those who want money from you. If they are taking a percentage, it shouldn't cost you anything. All my cover art and editing are free. Well, not really. They take a percentage of my royalty, but it isn't a big percentage.

My only complaint is that they use Smashwords (soon to be D2D) and don't publish directly to iTunes (books), Barnes & Noble, or others. But that would create a lot more headache for them in terms of accounting.
