I'm in the middle of a story and it needs a part where a new Domme is asked for a challenging scene by a more experienced malesub. I don't mean technically tricky, like knot tying, but something more psychologically unusual, that would push her out of her comfort zone. @Kumquatqueen - thanks for your list, but the story already has D/s 24/7 and some borderline non-con elements in it that cover off some of the other suggestions. I'm looking for quirky left-field that would fit into a single scene.

So I'd go with @joy_of_cooking 's ideas of her (or him) wanting something vanilla or emotional, to contrast with the kinks and provide a resolution to the story.

Humiliation and degradation, whether verbal or physical, could be something that a domme would be uncomfortable with. Some degrading name-calling, maybe even trying to undermine the sub's sense of self-worth could be the psychological aspect, and if you want to add a physical aspect too, she could treat the sub as an object, as furniture, for example, and wouldn't even use the sub's name.

Related forms of play are broader ethnic play, religion or political play, age play--say a younger Domme humiliating an older man for his aging, something I've written. Oh, and one of my favorites, class play: princess vs pauper. Never gone there, but disability play? In any case, I would think along those lines.

Second edit: Another button pushing direction, consider a forcefully face-riding menstruating Domme. Or something I do not believe fits with TFC "in real life" but might be a pretend scenario, pregnancy centered Dommeing.
