When our 2nd (and 3rd and 4th...) I could tell in a week or so, certainly by two weeks, because she felt different when fingering her vagina. I suspect it had to do with a mucus plug forming in her cervix. There were also other subtle changes, nothing specific, but general subtle differences from the overall way she felt, smelled, lubed, etc.
